Product Specific Information

The 16G6 antibody reacts with mouse Granzyme B (GrB), a member of the granzyme serine protease family found in cytotoxic T cells and NK cells. Granzyme B (also known as CGL1 or CTLA-1) is crucial for inducing apoptosis in target cells via interaction with cytotoxic T cells. The mannose 6-phosphate receptor acts as a death receptor for Granzyme B during this process.

For intracellular staining and flow cytometric analysis using direct conjugates of anti-mouse Granzyme B, it is recommended to use the Foxp3 Staining Buffer Set (Product # 00-5523).

Applications Reported:
The 16G6 antibody has been used for western blotting, immunohistochemical staining, ELISA capture, and staining of formalin-fixed paraffin-embedded tissue sections.

Applications Tested:

• Sandwich ELISA for mouse Granzyme B quantitation (capture antibody, 1–4 µg/mL)
• Western blotting (recommended starting concentration: 1 µg/mL)
• Formalin-fixed paraffin-embedded tissue (≤20 µg/mL, low pH antigen retrieval)

Careful titration is recommended for optimal assay performance.

Target Information

Granzyme B is a serine protease found in cytotoxic T cells and NK cells. It induces apoptosis through caspase-3 activation and mitochondrial pathways (via Bid cleavage). Granzymes (A–G) are stored in lytic granules of CTLs and NK cells. Granzyme B is essential for immune-mediated cytotoxicity and apoptosis induction.
