Product Specific Information

The monoclonal antibody HM104 recognizes the extracellular part of the Tumor Necrosis Factor Receptor type I (TNF-RI) of both membrane-bound and soluble forms. TNF-RI (~55–60 kDa) is expressed on most cell types and plays a key role in TNF-alpha–mediated cell stimulation. TNF-alpha triggers inflammatory responses, apoptosis, cellular proliferation, and may contribute to cancer progression. These effects are mediated through TNF-RI and TNF-RII, which share overlapping but distinct signaling pathways.

TNF-RI is activated by both soluble (17 kDa) and membrane-bound (26 kDa) TNF-alpha, while TNF-RII responds mainly to the membrane-bound form. Upon ligand binding, TNF-RI trimerizes and recruits a signaling complex including TRADD, RIP, cIAP1, TRAF2, and possibly TRAF5. Studies with TNF-RI-deficient mice demonstrate its major role in proliferation, inflammation, and apoptosis signaling.


Target Information

TNFR1 (Tumor Necrosis Factor Receptor 1) is a member of the TNF receptor superfamily and a principal receptor for TNF-alpha. It mediates cytokine signaling, activates NF-κB, promotes angiogenesis, and regulates gene expression. The extracellular domain can be released as soluble TNFR1 (sTNFR1). In humans, the TNFR1 gene is located on chromosome 12.

TNFR1 interacts with BAG4/SODD, TRADD, and TRAF2 proteins, which regulate downstream signaling. Mutations in TNFR1 extracellular domains are associated with autosomal dominant periodic fever syndrome, likely due to impaired receptor clearance.
