Description:
The monoclonal antibody eBio28F12 reacts with and inhibits the bioactivity of mouse TSLP.
Thymic Stromal-Derived Lymphopoietin (TSLP) was first identified in thymic stromal cell culture supernatant as a growth factor for pre-B cell proliferation and differentiation. It is structurally and functionally related to IL-7 and induces dendritic cells to promote Th2 differentiation and regulatory T cell development.
TSLP expression is induced by inflammatory cytokines and TLR ligands, regulated by NF-κB, and detected in epithelial, stromal, and basophil cells. Elevated TSLP levels are linked to asthma and atopic dermatitis, both Th2-mediated conditions. Chronic overexpression may increase allergen sensitivity.

The monoclonal antibody eBio28F12 reacts with and inhibits mouse TSLP bioactivity.
Applications Tested:
The ND₅₀ of eBio28F12, determined by neutralization of recombinant mouse TSLP-induced proliferation of the Nag8/7 cell line, is 1–3 ng/mL in the presence of 0.5 ng/mL mTSLP. The neutralization dose varies depending on cell type, assay method, and cytokine concentration.

Target Information
TSLP is a 159 amino acid four-helix bundle hematopoietic cytokine that signals through a heterodimeric receptor complex of the TSLP receptor and IL-7Rα. It affects myeloid cells, induces chemokine release from monocytes, enhances dendritic cell maturation, and promotes differentiation of naive CD4⁺ and CD8⁺ T cells into proallergic effectors.
TSLP primes dendritic cells to produce IL-12 following CD40 ligand stimulation, influences regulatory T cell selection, and maintains peripheral CD4⁺ T cell homeostasis. It can induce allergic inflammation via mast cell activation and may play a role in inflammatory arthritis. Expression is found in heart, liver, and prostate tissues.
