The monoclonal antibody eBioTH6 recognizes FR4, also known as FR delta and FBP3 (folate binding protein 3). FR4 is a heavily glycosylated 35 kDa receptor for folic acid and the physiologic circulating form of the vitamin, N5-methyltetrahydrofolate.
Natural regulatory T cells (Tregs) express high levels of FR4, which, together with CD25, distinguishes natural Tregs from effector, memory-like, and naïve T cells.

In vivo studies using eBioTH6 have demonstrated a reduction in CD4⁺CD25⁺ cells and enhanced tumor responses leading to elimination of advanced tumors.
Co-staining studies indicate that eBioTH6 and eBio12A5 recognize different epitopes, allowing for complementary functional analyses.

Applications Reported: Flow cytometric analysis, immunoprecipitation, and in vivo depletion.
Applications Tested: Tested by flow cytometric analysis of mouse splenocytes at ≤0.25 µg/test. A test is defined as the amount (µg) of antibody staining a cell sample in 100 µL. Optimal titration is recommended for each assay.

Filtration: 0.2 µm post-manufacturing filtered.
Purity: >90% (SDS-PAGE).

Target Information

FR4 is a 35 kDa glycoprotein receptor for folic acid and N5-methyltetrahydrofolate.
It is highly expressed on natural Tregs and used with CD25 to differentiate T cell subsets.
In vivo, eBioTH6 reduces CD4⁺CD25⁺ populations and enhances antitumor responses, leading to tumor elimination.
