Product Specific Information
The 6C2 monoclonal antibody reacts with mouse Toll-like receptor 2 (TLR2), expressed by the myeloid lineage including macrophages and dendritic cells in splenocytes and RAW264.7 cells. TLR2 and TLR4 are pattern recognition receptors involved in innate immunity and inflammation, recognizing bacterial lipoproteins.
Applications Reported:
6C2 is suitable for flow cytometric analysis. Preliminary studies suggest partial inhibition of monocyte activation through TLR2. For optimal staining, use directly conjugated 6C2 (FITC, PE, or Biotin) with FcR blocking using anti-mouse CD16/CD32 (clone 93, Product #14-0161).
Applications Tested:
Tested by flow cytometric analysis of mouse splenocytes or RAW264.7 cells. Recommended ≤1 µg per test (100 µL final volume). Cell number should be optimized empirically (10⁵–10⁸ cells/test). Titrate for best performance.

Target Information
TLR2 is a member of the Toll-like receptor family, crucial for pathogen recognition and activation of innate immunity. It interacts with adaptor molecules such as MyD88 and TIRAP to activate kinases and transcription factors (PKCα/β, NF-κB). TLR2 forms heterodimers with TLR1 or TLR6 to recognize bacterial and mycoplasma lipoproteins.
TLR2 is highly conserved across species and expressed predominantly in peripheral blood leukocytes. It mediates host responses to Gram-positive bacteria and yeast via NF-κB activation. Expression is regulated by immune modifiers: upregulated by LPS, GM-CSF, IL-1, IL-10; downregulated by IL-4, IFN-γ, TNF.
