Product Specific Information

Description:
The TLR3.7 monoclonal antibody reacts with human Toll-like receptor 3 (TLR3). This receptor family includes at least twelve members characterized by extracellular leucine-rich repeats and cytoplasmic domains homologous to the IL-1 receptor. TLR3 recognizes double-stranded RNA (dsRNA), activating NF-kappaB through MyD88-dependent and -independent pathways, and inducing type I interferons (IFNs). TLR3.7 suppresses poly(I):poly(C)-mediated IFN-beta production by human fibroblasts expressing TLR3.

Applications Reported:
Used in flow cytometric analysis and in vitro inhibition of ligand binding. For optimal flow cytometry detection, PE- or Biotin-conjugated TLR3.7 antibodies are recommended.

Applications Tested:
Tested by intracellular staining and flow cytometric analysis of A549 and MRC5 cell lines. Recommended usage ≤ 2 µg/test (100 µL final volume). Cell count per test may vary between 10⁵–10⁸ cells. Antibody titration is advised for optimal performance.

Storage and Handling:
Use in a sterile environment.

0.2 µm post-manufacturing filtered.

90%, determined by SDS-PAGE.

Target Information

TLR3 (Toll-like receptor 3, CD283) is a member of the TLR family involved in pathogen recognition and innate immunity activation. These receptors detect pathogen-associated molecular patterns (PAMPs) and mediate cytokine production for immune responses.
TLR3 is highly expressed in placenta and pancreas, mainly in dendritic cells. It recognizes viral dsRNA, activates NF-kappaB, and induces type I interferons. TLR3 signaling utilizes TRIF instead of MyD88, leading to IRF-3 and NF-kB activation. This pathway is crucial for antiviral defense and dendritic cell maturation.
TLR3 has been implicated in recognition of viruses such as Influenza A and West Nile Virus and may facilitate viral entry.
