Product Specific Information
The FF3399 monoclonal antibody recognizes human aryl hydrocarbon receptor (AHR), a ligand-activated transcription factor mediating the toxic effects of environmental contaminants such as PCBs and TCDD. AHR also binds naturally occurring compounds and is localized in the cytoplasm with HSP90, p23, and XAP2/AIP/ARA9. Upon ligand binding, AHR translocates to the nucleus, forming a complex with ARNT to regulate gene expression (e.g., CYP1A1). It is highly expressed in the liver and plays a role in Treg and Th17 cell regulation.

- Applications Tested: Pre-titrated and tested with Th17-polarized human PBMCs using Intracellular Fixation & Permeabilization Buffer Set (Product #88-8824-00) and Foxp3/Transcription Factor Staining Buffer Set (Product #00-5523-00).

Target Information
AHR (Ah Receptor) is part of a protein family including ARNT (HIF-1 Beta) and Drosophila PER/SIM proteins. It contains an N-terminal PAS domain (~200 aa) and binds DNA enhancer sequences to initiate transcription of cytochrome P450 (CYPIA1). AHR regulates xenobiotic-metabolizing enzymes and binds aromatic hydrocarbon ligands such as TCDD. In inactive form, AHR is cytoplasmic and complexed with HSP90; upon ligand binding, it translocates to the nucleus and forms a heterodimer with ARNT. Diseases associated with AHR include eosinophilic fasciitis and seborrheic dermatitis.
