Target Information
Tau는 신경세포 축삭에서 주로 발견되는 미세소관 결합 단백질로, 튜불린 중합을 촉진하고 미세소관을 안정화시킵니다.
C-말단은 축삭 미세소관에 결합하고 N-말단은 신경 세포막 구성 요소에 결합하여, Tau가 두 구조 사이를 연결하는 역할을 합니다.
TAU/MAPT의 세포 내 위치는 축삭 극성을 결정하며, 짧은 isoform은 세포골격의 유연성을, 긴 isoform은 안정성을 담당합니다.
과인산화된 형태의 Tau는 Alzheimer’s disease (AD) 뇌의 paired helical filaments(PHF)의 주요 구성 요소로, 미세소관 결합 기능을 저하시켜 신경세포 퇴화를 유발합니다.
다양한 serine/threonine kinase(GSK-3β, PKA, cdk5, casein kinase II 등)가 Tau를 인산화하며, Pick’s disease, frontotemporal dementia, cortico-basal degeneration, progressive supranuclear palsy 등 여러 신경퇴행성 질환에서 과인산화된 Tau가 발견됩니다.
