Product Specific Information

The monoclonal antibody 1H9 recognizes both mouse and human Pax5, also known as BSAP (B cell specific activator protein). Pax5 is a member of the paired box (pax) family of transcription factors and the only member expressed in hematopoietic cells.

During embryogenesis, Pax5 is expressed in the brain of mice and in the mesencephalon and spinal cord of humans. Its expression is upregulated early in B cell development and maintained through most subsequent stages. Suppression of Pax5 is essential for plasma cell differentiation via Blimp-1 expression.

In the spleen, Pax5 expression is higher in marginal zone B cells (B220+ CD21^high CD23^low) than in other B cells. Pax5 also influences VH-DJH recombination and regulates various B and non-B cell proteins. Mutations or altered expression of Pax5 are associated with several neoplasms such as DLBCL, non-Hodgkin lymphomas, and B-cell ALL. Translocations with elastin, IGH, ETV6, FOXP1, and EVI3 have been reported.

• Recommended use ≤ 0.25 µg/test (100 µL final volume, 10⁵–10⁸ cells/test).
• Optimize antibody titration for best performance.

Target Information

PAX5 is a member of the paired box (PAX) family of transcription factors that regulate early development. PAX5 (BSAP) is expressed in early B-cell differentiation but not in late stages. It is also expressed in developing CNS and testis, suggesting a role in neural development and spermatogenesis. Mutations in PAX5 are linked to leukemia and acute lymphoblastic conditions.
