Product Specific Information

The monoclonal antibody TWAJ recognizes mouse and human Gata-3, a T cell-specific transcription factor critical for thymic development and Th2 differentiation.
Gata-3 expression is also found in the CNS, skin, mammary glands, and kidney during embryonic development. Homozygous Gata-3 knockout is embryonic lethal, indicating its essential role.

This antibody recognizes both 50 kDa and 45 kDa forms of Gata-3.
Staining requires the Foxp3/Transcription Factor Staining Buffer Set (Product #00-5523-00).
Cross-reactivity in rhesus monkeys has been reported.

Applications Reported: Intracellular staining followed by flow cytometric analysis.
Applications Tested: Pre-titrated and validated for intracellular staining of mouse thymocytes using the Foxp3/Transcription Factor Staining Buffer Set (Product #00-5523).

Use at 5 µL (0.00375 µg) per test (100 µL final volume). Cell number may vary from 10⁵–10⁸ cells/test.

Light Sensitivity: Protect from light; tandem dye is sensitive to photo-induced oxidation.
Fixation: Samples can be stored in IC Fixation Buffer (Product #00-8222) or 1-step Fix/Lyse Solution (Product #00-5333) for up to 3 days at 4°C in the dark.
Excitation: 488–561 nm
Emission: 775 nm
Laser: Blue, Green, Yellow-Green
Filtration: 0.2 µm post-manufacturing filtered.

Target Information

GATA3 is a zinc finger transcription factor that binds enhancer elements of all four TCR genes (α, β, γ, δ), regulating T-cell receptor gene activation.
It is expressed in T cells but not in B cells or macrophages.
The human gene spans 17 kb with six exons, encoding two zinc fingers highly conserved with GATA1.
