Description:
The dG9 antibody clone reacts with human perforin (pore-forming protein, pfp). Perforin is a cytolytic mediator found in cytotoxic T lymphocytes (CTL) and natural killer (NK) cells, playing a key role in immune defense against tumors and viral infections.

Applications Reported:

• Intracellular staining followed by flow cytometric analysis.

Applications Tested:

• Pre-titrated and validated for intracellular staining and flow cytometric analysis of normal human peripheral blood cells.
• Recommended use: 5 µL (0.5 µg) per test (100 µL final volume; 10⁵–10⁸ cells/test).

Light Sensitivity:

• This tandem dye is sensitive to photo-induced oxidation. Protect from light.

Fixation:

• Samples can be stored in:
  • IC Fixation Buffer (Product # 00-8222-49)
  • 1-step Fix/Lyse Solution (Product # 00-5333-54)
    for up to 3 days at 4°C in the dark with minimal impact on brightness and FRET efficiency.

Target Information

Perforin is a major cytolytic protein stored in cytoplasmic granules of cytotoxic lymphocytes. It mediates immune defense against tumors and viral infections through pore formation in target cell membranes.

• Protein size: 555 amino acids (70–75 kDa)
• Mechanism: Transmembrane channel formation similar to complement component C9
• Expression: Limited to killer cell lines (not in helper T cells or other tumor cells)
• Clinical relevance: Mutations in the perforin gene cause familial hemophagocytic lymphohistiocytosis type 2 (HPLH2), a rare autosomal recessive disorder.
• Alternative splicing: Multiple transcript variants exist.
