Product Specific Information

Description:
The LN3 monoclonal antibody reacts with human MHC class II molecule HLA-DR, expressed on antigen-presenting cells (B cells, monocytes, macrophages, dendritic cells, and activated T cells). HLA-DR is a heterodimeric transmembrane protein essential for peptide presentation to CD4⁺ T lymphocytes.

Applications Reported:
Validated for flow cytometric analysis.

Applications Tested:
Pre-titrated and tested using normal human peripheral blood cells. Use 5 µL (0.015 µg) per test (final volume 100 µL, 10⁵–10⁸ cells/test).

Light Sensitivity:
Protect from light; tandem dye is sensitive to photo-induced oxidation.

Fixation:
Samples may be stored in:

Excitation / Emission / Laser:
488–561 nm / 775 nm; Blue, Green, Yellow-Green lasers.

Filtration:
0.2 µm post-manufacturing filtered.


Target Information

HLA-DR is a transmembrane glycoprotein composed of a 36 kDa alpha chain (DRA) and a 27 kDa beta chain (DRB).

  • Alpha chain (DRA): Non-polymorphic, acts as sole alpha chain for DRB1, DRB3, DRB4, DRB5.
  • Beta chain (DRB): Contains polymorphisms determining peptide-binding specificity.
  • Expression: Primarily on antigen-presenting cells (B lymphocytes, monocytes, macrophages, thymic epithelial cells, activated T lymphocytes).
  • Function: Binds processed peptides, presents to T-helper cells, critical for immune response initiation.
  • Applications: Widely used in immunopeptidomics for neoantigen identification via LC-MS/MS.
