Description:
The monoclonal antibody H17F10A7 reacts with human IL-17F, a member of the IL-17 family of pro-inflammatory cytokines. Like IL-17A, IL-17F is a disulfide-linked, homodimeric glycoprotein containing a classical cysteine knot motif, also found in TGF-β, BMP, and NGF superfamilies. This motif suggests potential heterodimeric structures, as seen in TGF-β and inhibin/activin.

Recent studies show that co-expression of IL-17F and IL-17A in HEK293 cells forms biologically active IL-17F/IL-17A heterodimers, in addition to IL-17F and IL-17A homodimers. Activated human CD4+ T cells also produce IL-17A/F heterodimers.
Comparative studies indicate that IL-17A, IL-17A/F, and IL-17F all induce GRO-α secretion, with IL-17A being the most potent, followed by IL-17A/F, and IL-17F (100-fold lower activity than IL-17A).
These heterodimers can be detected via immunoprecipitation using eBio64CAP17 anti-IL-17A monoclonal antibody followed by immunoblotting with H17F10A7.

Target Information

IL-17F (Interleukin 17F, CTLA-8) is a cytokine produced by inflammatory cells such as activated T cells, mast cells, and basophils. It is involved in allergic airway inflammation and induces cytokines, chemokines, and adhesion molecules in epithelial, endothelial, and fibroblast cells.

IL-17F may exist as a homodimer or heterodimer with IL-17A, acting through IL-17R to recruit monocytes and neutrophils by enhancing chemokine production. It also stimulates TNF-α, IL-1β, IL-6, and IL-8 production and is implicated in chronic inflammatory diseases such as rheumatoid arthritis, psoriasis, and multiple sclerosis.

TGF-β (for differentiation) and IL-23 (for expansion) are required for Th17 cell induction and maintenance. IL-17F is homodimeric, homologous to a herpes virus early protein, and one of six IL-17 family members (IL-17A–F). It is highly expressed by activated effector memory T cells and can inhibit endothelial angiogenesis while inducing IL2, TGFB1, and MCP-1 production.
