Product Specific Information
The MP6-XT22 antibody reacts with mouse tumor necrosis factor-alpha (TNF alpha), a 17 kDa cytokine produced by monocytes, macrophages, neutrophils, NK cells, and CD4(+) T cells. TNF alpha has cytolytic activity against various tumor cells and plays an important role in immune regulation. It exists in soluble (17 kDa) and membrane-bound (26 kDa) forms, forming dimers and trimers.
Applications Reported:
- Intracellular staining followed by flow cytometric analysis
- Immunohistology staining of frozen tissue sections
(Fluorochrome-conjugated MP6-XT22 is recommended for intracellular flow cytometry.)
Applications Tested:
- Tested to block staining with fluorochrome-conjugated MP6-XT22
- Recommended ≤ 2 µg/test (in 100 µL final volume)
- Optimal titration required for specific assays
- Suitable for 10⁵–10⁸ cells/test
Purity: >90% (SDS-PAGE)
Aggregation: <10% (HPLC)
Filtration: 0.2 µm post-manufacturing filtered
Target Information
TNF alpha is a multifunctional proinflammatory cytokine belonging to the TNF superfamily. It is mainly secreted by macrophages and binds to receptors TNFRSF1A/TNFR1 and TNFRSF1B/TNFBR. TNF alpha regulates immune cell function, proliferation, differentiation, apoptosis, lipid metabolism, and coagulation. It forms multimers (dimers, trimers, pentamers) and shows a single 17 kDa band under non-reducing SDS-PAGE conditions.
Knockout mouse studies indicate TNF alpha’s neuroprotective role and tumor necrosis induction in tumor-bearing mice. It also contributes to immune responses against bacterial, viral, parasitic, and fungal infections and participates in tumor cytotoxicity, synergistic with IFN-gamma. TNF alpha is implicated in autoimmune diseases, insulin resistance, and cancer.
