Target Information
This gene encodes a G protein-coupled receptor with selectivity for three chemokines: IP10 (interferon-g-inducible 10 kDa protein), Mig (monokine induced by interferon-g), and I-TAC (interferon-inducible T cell a-chemoattractant).
These chemokines belong to the CXC subfamily, characterized by a single amino acid residue separating the first two of four conserved cysteine residues.
Binding of chemokines to CXCR3 induces cellular responses such as leukocyte trafficking, integrin activation, cytoskeletal reorganization, and chemotactic migration. Inhibition by Bordetella pertussis toxin suggests coupling to Gi-subclass G proteins.
CXCR3 is prominently expressed in effector/memory T cells and in T cells within inflamed tissues. Its ligands IP10, Mig, and I-TAC are produced locally in inflammatory lesions, implicating CXCR3 in inflammatory cell recruitment.
This receptor is a potential target for small-molecule antagonists aimed at treating inflammatory diseases. Multiple transcript variants encoding different isoforms have been identified.
