Product Specific Information
The monoclonal antibody DECMA-1 recognizes mouse, human, and canine CD324 (E-Cadherin, also known as epithelial cadherin or uvomorulin). E-Cadherin is a transmembrane glycoprotein involved in intercellular adhesion, sharing structural features with other cadherin family members (P and N cadherin). Its extracellular domain contains repeating Ca²⁺-binding motifs, while the cytoplasmic domain interacts with catenins (α, β, γ) and actinins, linking E-Cadherin to the cytoskeleton.
E-Cadherin is expressed in most epidermal cells, including melanocytes and keratinocytes, and localized at epithelial cell boundaries. It plays a critical role in maintaining tissue integrity. Loss of E-Cadherin function is associated with cancer progression.
Note: E-Cadherin is sensitive to trypsin; avoid trypsin exposure.
The DECMA-1 antibody demonstrates functional activity by disrupting adhesion in human, mouse, and dog cells.
Applications Reported: Flow cytometry, Western blotting, and immunohistochemistry (frozen tissue).
Applications Tested: Flow cytometric analysis of MDCK cells. Optimal staining achieved by intracellular staining due to variable surface expression. Recommended ≤0.25 µg/test (100 µL final volume, 10⁵–10⁸ cells/test). Antibody titration is advised for optimal assay performance.
Target Information
E-Cadherin (epithelial cadherin) is a classical cadherin from the calcium-dependent adhesion protein superfamily. It is a cell-cell adhesion glycoprotein with five extracellular cadherin repeats, a transmembrane region, and a conserved cytoplasmic tail. Mutations are linked to gastric, breast, colorectal, thyroid, and ovarian cancers. Loss of E-Cadherin function promotes tumor invasion and metastasis. The ectodomain mediates bacterial adhesion, while the cytoplasmic domain enables internalization. The human CDH1 gene on chromosome 16 encodes E-Cadherin.
E-Cadherin is essential for tissue architecture and integrity maintenance. Reduced expression correlates with increased tumor invasiveness.
