Product Specific Information
Description:
The monoclonal antibody 29A1.4 recognizes mouse NKp46 (CD335), a member of the natural cytotoxicity receptor (NCR) family. NKp46 is a glycoprotein with two Ig-like domains and a short cytoplasmic tail, expressed specifically on NK cells (including immature NK cells, DX5⁻ CD3⁻). This enables discrimination between NKT and NK cells (NKp46⁺, CD3⁻).
Unlike other NK markers, staining with 29A1.4 is not strain-specific and has been validated in C57Bl/6, SJL, CBA/CA, and BALB/C strains. NKp46 participates in NK cell-mediated lysis of tumor and pathogen-infected cells.
The 29A1.4 antibody activates NK cells in vitro but does not deplete them in vivo.
Applications Reported: Flow cytometric analysis.
Applications Tested: Tested by flow cytometry on mouse splenocytes. Recommended ≤0.125 µg/test.
A test corresponds to the antibody amount for staining a cell sample in 100 µL final volume (10⁵–10⁸ cells/test). Optimize titration for best results.
Filtration: 0.2 µm post-manufacturing filtered.
Target Information
NKp46 (CD335, NCR1) is a cytotoxicity-activating receptor that enhances NK cell-mediated tumor cell lysis. It contains two Ig-like domains related to leukocyte immunoglobulin-like (LIR) and killer inhibitory receptors (KIR).
