Product Specific Information
The HTA28 monoclonal antibody recognizes phosphorylated serine 28 of histone H3 in human, mouse, rat, bovine, and hamster. Histone H3 (15 kDa) is a chromatin component forming nucleosome structure. Phosphorylation at Ser28 is mediated by MSK1 following MAP kinase activation in response to tumor promoters (UV, EGF) and oncoproteins (c-Myc, c-Jun, c-Fos). This modification is associated with chromosome condensation during mitosis, cell transformation, and RNA polymerase III transcription regulation.
Applications Reported: Flow cytometric analysis.
Applications Tested: Pre-titrated for intracellular staining of nocodazole-treated HeLa cells (≤ 0.5 µg/test). A test is defined as the antibody amount for staining a 100 µL cell sample (10⁵–10⁸ cells/test). Optimize titration for best results.
Recommended Protocols:
- Protocol A: Two-step (intracellular/cytoplasmic proteins)
- Protocol C: Two-step (Fixation/Methanol)
- Protocol B (one-step, nuclear proteins) cannot be used.
