Target Information
EIF2AK2 (PKR) is one of four kinases that phosphorylate Ser51 of translation initiation factor eIF2-alpha in response to environmental stresses. PKR plays a key role in antiviral defense, apoptosis, and cell growth regulation. It mediates IFN-induced antiviral response and virus-induced apoptosis through Fas expression and FADD/Caspase-8 signaling. PKR also regulates NF-κB, p53, p38, and PDGF pathways, and is implicated in tumor suppression and neurodegenerative diseases such as Alzheimer’s, Huntington’s, and ALS. Viruses can inhibit PKR activity, e.g., by influenza virus NS1 protein.
