Target Information
This gene encodes a transmembrane receptor commonly referred to as DC-SIGN, expressed on the surface of dendritic cells and macrophages. It plays a critical role in the innate immune system by recognizing a wide range of pathogens, including parasites and viruses.
The protein has three main domains:
- N-terminal transmembrane domain
- Tandem-repeat neck domain
- C-type lectin carbohydrate recognition domain
The extracellular region (neck + lectin domains) functions both as a pathogen recognition receptor and a cell adhesion receptor, binding carbohydrate ligands on microbial and host cell surfaces. The neck region mediates homo-oligomerization, enhancing multivalent ligand binding. Variations in the 23-amino-acid repeats within this region can affect binding affinity.
DC-SIGN is closely related to L-SIGN (GeneID 10332), sharing sequence and functional similarities but differing in ligand specificity and tissue distribution. Alternative splicing leads to multiple isoforms.
