Product Specific Information
The Clone 9 monoclonal antibody reacts with mouse B7S1, also known as B7H4 — a newly discovered member of the B7 family. B7S1 is thought to regulate T-cell costimulation influenced by B-cell activation status. The ligand for B7S1 has not been identified, and this antibody is non-blocking.
Double staining with anti-mouse B7-H4 antibodies Clone 9 and 188 suggests that their epitopes differ or do not interfere sterically when used together.
Applications Reported: Suitable for flow cytometric analysis.
Applications Tested: Tested on transfected cells; ≤0.5 µg per test (100 µL final volume). Cell number may vary between 10⁵–10⁸ cells/test. Antibody titration recommended for optimal performance.

Target Information
B7-H4 (B7x, VTCN1) is a costimulatory protein functioning as a negative regulator of T-cell mediated immunity. Although its receptor is unknown, B7-H4 delivers inhibitory signals to T-cells, suppressing proliferation, cell cycle progression, and IL-2 production.
B7-H4 deficient mice show minimal immune alteration, suggesting its role in fine-tuning T-cell responses. It is expressed on activated T-cells, B-cells, monocytes, and dendritic cells. Abnormal expression is linked to lung, breast, and ovarian cancers.
B7-H4 primarily localizes on lymphoid cell membranes and interacts with T lymphocyte receptors to modulate immune responses. Overexpression may help tumor cells evade immune detection and facilitate progression.
VTCN1 may serve as a biomarker for early detection of ovarian cancer and renal cell carcinoma (RCC).
