The 1A29 monoclonal antibody reacts with rat CD54, also known as intercellular adhesion molecule 1 (ICAM-1).
This type I immunoglobulin-like transmembrane protein is expressed on lymphocytes, macrophages, dendritic cells, monocytes, granulocytes, and thymic stromal cells, as well as endothelial and non-immune cells such as hepatocytes and cardiomyocytes.

CD54 expression can be induced in vitro by proinflammatory cytokines (IFN-γ, IL-1, CCL2, TNF-α). It plays a key role in transendothelial migration of leukocytes and T cell interactions with antigen-presenting cells, mediated via binding to LFA-1 (CD11a/CD18).
The 1A29 antibody has been reported to block interactions between CD54 and LFA-1.

Applications Reported: Flow cytometric analysis
Applications Tested: Tested on rat splenocytes at ≤0.125 µg/test (100 µL final volume, 10⁵–10⁸ cells/test). Antibody titration recommended for optimal performance.

Fluorophore Information:
PerCP-eFluor® 710 emits at 710 nm and is excited by the blue laser (488 nm).
Recommended filter: 710/50 bandpass (alternative: 695/40 bandpass). Ensure instrument compatibility.

Fixation:
Samples can be stored in IC Fixation Buffer (Product #00-8222) or 1-step Fix/Lyse Solution (Product #00-5333) for up to 3 days at 4°C in the dark. Clone-specific performance should be determined empirically.

Target Information

ICAM-1 (CD54) is an 85–110 kDa single-chain type I integral membrane glycoprotein composed of five immunoglobulin superfamily repeats, a transmembrane region, and a cytoplasmic domain.
It has seven potential N-linked glycosylation sites and shares sequence homology with ICAM-2 (CD102) and ICAM-3 (CD50).
ICAM-1 binds to integrins CD11a/CD18 (LFA-1) and CD11b/CD18 (Mac-1), and serves as a receptor for Rhinovirus.

Expressed by activated endothelial cells and detected on epithelial cells, fibroblasts, chondrocytes, B and T lymphocytes, monocytes, macrophages, dendritic cells, and neutrophils. Expression increases upon inflammation.
Soluble ICAM-1 is detectable in plasma and elevated in various inflammatory conditions.
