Product Specific Information

The HM48-1 monoclonal antibody reacts with the mouse CD48 antigen (also known as BCM1, Blast-1 in human, and OX-45 in rat). CD48 is a 45 kDa GPI-linked glycoprotein expressed on most hematopoietic cells and belongs to the SLAM family.
It is a useful marker for distinguishing hematopoietic progenitor subsets based on SLAM family expression patterns (CD150, CD244, CD48).

• Restricted progenitors: CD48(+)CD244(+)CD150(-)

CD48 plays a key role in adhesion and T-cell activation. In mice, CD2 and CD244 serve as its primary counter-receptors. HM48-1 can modulate CD48 functions in vitro and in vivo, including blocking CD48/CD2 and CD48/CD244 interactions, inhibiting mitogen-induced proliferation, providing T-cell costimulation, and prolonging cardiac allograft survival.

Applications Reported: Flow cytometric analysis
Applications Tested: Tested on mouse splenocytes at ≤0.125 µg/test (100 µL final volume, 10⁵–10⁸ cells/test). Optimal titration recommended.

Fluorochrome Information:
PerCP-eFluor® 710 emits at 710 nm and is excited by the 488 nm blue laser. Recommended filter: 710/50 bandpass (695/40 acceptable alternative). Ensure instrument compatibility.

Fixation:
Samples can be stored up to 3 days at 4°C in the dark using:

• IC Fixation Buffer (Product #00-8222-49)
• 1-step Fix/Lyse Solution (Product #00-5333-54)

Target Information

CD48 (Blast-1) is a GPI-anchored member of the CD2 subset of the Ig superfamily, which includes CD2, CD58, CD84, CD150, CD229, and CD244. These molecules mediate homotypic or heterotypic adhesion through Ig family interactions.

• Broadly expressed on hematopoietic cells
• High-affinity ligand for 2B4 and low-affinity ligand for CD2
• Regulates NK and NK-T cell proliferation via 2B4-CD48 interaction
• Involved in eosinophil activation and immune responses
• Strongly expressed on lymphocytes and monocytes, weakly on granulocytes
• Absent on platelets, fibroblasts, epithelium, and endothelium
• Marker for detecting GPI-anchor defects (e.g., PNH)
• Potential pharmacological target in allergic airway diseases (e.g., rhinitis, asthma)
