Description:
R1-2 단클론 항체는 마우스 CD49d (150 kDa integrin α4 서브유닛)와 반응합니다. CD49d는 integrin β1(CD29)과 비공유 결합하여 VLA-4 복합체를 형성하며, fibronectin 및 VCAM-1(CD106)의 수용체로 작용합니다. 또한 integrin β7과 결합하여 MAdCAM-1과 상호작용합니다. CD49d는 흉선세포, 말초림프구, 단핵구, 호산구에서 발현됩니다.

Applications Reported:
Flow Cytometry 분석에 사용 보고됨.

Applications Tested:
마우스 비장세포에서 Flow Cytometry로 테스트됨. 0.5 µg/test 이하 농도로 사용 권장. 최적의 성능을 위해 적정 농도는 실험적으로 결정 필요.

Fluorochrome Information:
PerCP-eFluor® 710은 488 nm(블루 레이저)로 여기되며, 710 nm에서 방출합니다. 710/50 또는 695/40 밴드패스 필터 사용 권장.

Fixation:
시료는 다음 조건에서 보관 가능:

• IC Fixation Buffer (Product #00-8222-49)
• 1-step Fix/Lyse Solution (Product #00-5333-54)
  4°C 암소에서 최대 3일간 보관 시 형광 세기 및 FRET 효율 유지.

Filtration:
제조 후 0.2 µm 필터링 완료.

Target Information

Integrin α4(CD49d, ITGA4)는 integrin α 체인 단백질군에 속하며, β1 또는 β7 체인과 결합하여 α4β1(VLA-4) 또는 α4β7 복합체를 형성합니다. 이 복합체는 fibronectin, VCAM, MadCAM 등과 결합하며, 특히 MadCAM은 α4β7의 주요 리간드로 알려져 있습니다. 또한 HIV-1 envelope 단백질이 MadCAM을 모방하여 α4β7을 통해 결합 및 신호전달을 유도할 수 있음이 보고되었습니다.
