Product Specific Information

• Specificity: A54 binds specifically to the D1 domain of CD40. It can only bind to the intact (non-truncated) version of CD40 and when an AD2 peptide tag has not been added. Recognizes CD40 on B cell line BJAB.
• Preparation: Selected from the n-CoDeR phage display library after 4 rounds of selection using biotinylated CD40-Fcgamma fusion protein as bait and capture on streptavidin-coated magnetic beads.

Target Information

CD40 antigen is a single-chain glycoprotein, a member of the tumor necrosis factor/nerve growth factor superfamily, showing significant homology to CD30. It is expressed on all B cells except plasma cells and also found on some epithelial cells, carcinomas, and lymphoid dendritic cells.
CD40 mediates immune and inflammatory responses including:

• T cell-dependent immunoglobulin class switching
• Memory B cell development
• Germinal center formation

The AT-hook transcription factor AKNA regulates CD40 expression, facilitating homotypic cell interactions. TNFR2 acts as an adaptor protein mediating CD40 signal transduction.
The CD40-CD40L interaction is crucial for amyloid-beta-induced microglial activation, implicated in early Alzheimer’s disease pathogenesis.
Two alternatively spliced transcript variants of CD40 encoding distinct isoforms have been reported.
Diseases associated with CD40 dysfunction include Type 3 Hyper-IgM immunodeficiency and CD40 ligand deficiency.
