Product Specific Information

• Specificity: ch5D12 binds specifically to recombinant and native human CD40 ectodomain and also binds CD40 in cynomolgus and marmoset monkeys. The antibody requires residues within D1 and D1/B2 for binding (D1 domain is crucial for CD40L binding).
• Preparation: Anti-CD40 mAb 5D12 was generated by immunizing mice with sf9 insect cells expressing recombinant human CD40 and selected for the ability to bind EBV-immortalized human B cells. The variable regions from mu5D12 were cloned and used to construct chimeric humanized IgG4 5D12.

Target Information

The CD40 antigen is a single chain glycoprotein belonging to the tumor necrosis factor/nerve growth factor superfamily and shows significant homology to the Hodgkin’s disease-associated antigen CD30.
CD40 is expressed on all B cells except plasma cells, and also found on epithelial cells, carcinomas, and lymphoid dendritic cells. It plays a crucial role in immune and inflammatory responses, including T cell-dependent immunoglobulin class switching, memory B cell development, and germinal center formation.
AT-hook transcription factor AKNA regulates CD40 expression, facilitating homotypic cell interactions. TNFR2 interacts with CD40 to mediate signal transduction.
The CD40-CD40L interaction is necessary for amyloid-beta-induced microglial activation, an early event in Alzheimer’s disease pathogenesis. Two alternatively spliced transcript variants encoding distinct isoforms have been reported.
Diseases associated with CD40 dysfunction include Type 3 Hyper-IgM immunodeficiency and CD40 ligand deficiency.
