Product Specific Information
The TY25 monoclonal antibody reacts with mouse B7-DC/PD-L2, a member of the B7 family (~25 kDa). B7-DC is expressed mainly by dendritic cells, monocytes, and macrophages. It binds PD-1 but not CD28/CTLA-4, mediating costimulation or suppression of T cell proliferation depending on activation state. TY25 is useful for studying B7-DC/PD-L2 function and expression in APC/T cell interactions.
Applications Reported:
TY25 has been used for flow cytometry, immunoprecipitation, Western blot (non-reducing conditions), and IHC of frozen sections. Also reported for ligand inhibition assays. Functional assays should use Functional Grade purified TY25 (Product #16-5986).
Applications Tested:
Tested by flow cytometry on mouse splenocytes and B7-DC transfected cells. Recommended ≤0.5 µg/test in 100 µL final volume. Cell number: 10⁵–10⁸ cells/test. Titrate for optimal performance.

Target Information
Programmed death-ligand 2 (PD-L2, B7-DC) is part of the B7 ligand family and binds PD-1. Expressed mainly in dendritic cells but inducible in other immune and non-immune cells. Expression is upregulated by IL-4 and TNF-α/IFN-γ. PD-L2 shows higher affinity to PD-1 than PD-L1 and functions as an inhibitory immune checkpoint molecule. It can also stimulate T cell proliferation even in PD-1-deficient cells, suggesting additional receptors such as RGMb. PD-L2–RGMb interaction regulates respiratory tolerance via BMP/neogenin signaling.
