Product Specific Information

The DX5 monoclonal antibody reacts with CD49b, an antigen expressed on most mouse natural killer (NK) cells and a subset of T cells. DX5 recognizes all tested mouse strains, including BALB/c, C57BL/6, C3H, CBA, DBA, AKR, SJL, and 129. Co-staining of C57BL/6 spleen cells with anti-NK1.1 (PK136) and DX5 shows coexpression in most cells, as well as distinct DX5⁺PK136⁻ and DX5⁻PK136⁺ populations.

Applications Reported: Flow cytometric analysis.
This antibody does not block NK cell adhesion or function and is not suitable for immunoprecipitation or immunohistochemistry of mouse tissues.

Applications Tested:
Tested by flow cytometry using mouse splenocytes. Recommended ≤0.5 µg per test (100 µL final volume). Optimal titration should be determined empirically (10⁵–10⁸ cells/test).

Target Information

CD49b (Integrin alpha 2) is a 150 kDa glycoprotein that pairs with CD29 (Integrin beta 1) to form the heterodimeric integrin VLA-2 (α2β1, GPIa/IIa). VLA-2 functions as a receptor for laminin, collagen, and fibronectin, mediating cell adhesion and signal transduction. CD49b is implicated in VEGF-induced angiogenesis and lymphocyte activation. It is expressed on NK cells, NK-T cells, monocytes, platelets, epithelial cells, and subsets of CD4⁺ T and B cells, including Tr1 regulatory T cells.
