Product Specific Information
Description:
The 12.14 monoclonal antibody reacts with mouse Nur77 (NR4A1, TR3, NGFI-B, NAK1), an inducible orphan nuclear receptor. Nur77 is expressed in thymocytes and T cell lines, promoting apoptosis and playing a role in thymocyte negative selection. It is also critical for steroid biosynthesis in Leydig cells and dopamine signaling. Nur77 interacts with FoxP3 in regulatory T cells, though results with this antibody do not correlate with that observation.

This antibody has been reported for use in immunoblotting (WB).
Applications Tested:
Tested by Western blot on mouse thymocyte lysates stimulated with PMA and ionomycin for 2 hours. Recommended concentration: 5 µg/mL (titrate for optimal performance).

Target Information
NR4A1 is a member of the steroid-thyroid hormone-retinoid receptor superfamily. It is expressed in brain, adrenal, thyroid, liver, testis, ovary, thymus, muscle, lung, and prostate. Expression is induced by stress stimuli and growth factors. Acts as a nuclear transcription factor mediating apoptotic signaling in thymocytes and tumor cells, and functions in hypothalamic-pituitary axis signaling. Translocation from nucleus to mitochondria induces apoptosis.
Three transcript variants encode two distinct isoforms of NR4A1. Variant (3), also known as TRC beta, has multiple differences from variant 1, resulting in isoform b with a shorter and distinct C-terminus. RXR is a known partner for NR4A1.
