ATG5 (Autophagy Related 5)는 자가포식 과정의 핵심 요소로, 세포 사멸 및 미토콘드리아 품질 관리, 선천적 항바이러스 면역 반응의 음성 조절, 림프구 발달 및 증식, MHC II 항원 제시, 지방세포 분화 등 다양한 세포 과정에 관여합니다.
ATG12와의 결합 후 자가포식소체 형성에 참여하며, FADD와 상호작용하여 자가포식성 세포 사멸에 기여합니다.
ATG5-ATG12 복합체는 막 형성 후 자가포식소체 완성 직전 분리되며, RIG-I 및 VISA(IPS-1) 단백질과 결합하여 type I interferon 생성을 억제하고 바이러스 복제를 허용합니다.
ATG5 기능 이상은 소뇌성 운동실조(spinocerebellar ataxia)와 연관되어 있습니다.
