- In Western blot, a band is observed around 56 kDa representing the ATG5-ATG12 complex (human ATG5 ≈ 33 kDa).
- Antigen retrieval with sodium citrate buffer (pH 6.0) is recommended prior to immunostaining paraffin tissues.
- Suggested positive control: mouse wildtype ES cell lysate.
Target Information
ATG5 (Autophagy Related 5) plays a critical role in autophagy and apoptotic processes. It is involved in:
- Mitochondrial quality control after oxidative damage
- Negative regulation of innate antiviral immune response
- Lymphocyte development and proliferation
- MHC II antigen presentation and adipocyte differentiation
Upon conjugation with ATG12, the complex contributes to autophagosome formation. ATG5 also interacts with FADD, influencing autophagic cell death. The ATG5-ATG12 conjugate associates with immune response proteins such as RIG-I and VISA, modulating type I interferon production and viral replication. Dysfunction of ATG5 is linked to diseases such as spinocerebellar ataxia.
