Interleukin 1 (IL1), originally known as lymphocyte activating factor (LAF), activates T cells and lymphocytes, which then proliferate and secrete interleukin 2. IL1 is primarily released from stimulated macrophages and monocytes, but also from several other cell types, and plays a key role in inflammatory and immune responses. The two closely related agents, interleukin-1 alpha (IL-1α) and interleukin-1 beta (IL-1β), bind to the same cell surface receptor, elicit nearly identical biological responses, and share 25% homology in their amino acid sequences.

Target Information

Interleukin-1 beta (IL-1β) is a proinflammatory cytokine expressed by monocytes, macrophages, and dendritic cells. It is synthesized as a 31 kDa inactive pro-form that accumulates in the cytosol and is cleaved into an active 17 kDa protein upon inflammasome activation. Inflammasomes are multi-protein complexes that respond to pathogens, stress, and danger signals, activating caspase-1, which in turn cleaves pro-IL-1β.

IL-1β lacks a signal peptide for the classical ER/Golgi pathway and is secreted via an alternate, incompletely understood mechanism. Although typically secreted in its active form, uncleaved IL-1β may also be detectable under some conditions. It signals through IL-1RI and IL-1RII receptors, shared with IL-1α. IL-1β activity can be regulated by IL-1 Receptor Antagonist (IL-1RA), which blocks receptor binding through competitive inhibition.

IL-1β plays a central role in innate host defense, triggering the production of other proinflammatory cytokines and initiating acute-phase responses to infection and injury. Elevated IL-1β levels are associated with various chronic inflammatory conditions, and IL-1β neutralizing antibodies have potential therapeutic value.
