Product Specific Information

The monoclonal antibody IgE21 recognizes the immunoglobulin E (IgE) class of antibodies. IgE levels in serum are naturally low and play a key role in allergic responses by binding to mast cells and basophils via the Fc epsilon receptor I, triggering histamine release. IgE is produced by plasma B cells and basophils, and exists in both soluble and membrane-bound forms. IgE21 recognizes both forms.

Each product contains:

• 1 vial of NovaFluor conjugate
• 1 vial of CellBlox Plus Blocking Buffer

Applications Reported: Suitable for flow cytometric analysis.
Applications Tested: Pre-titrated and validated on normal human peripheral blood cells. Recommended use is 5 µL (0.2 µg) per test in a final volume of 100 µL.

Note:

• NovaFluor dyes are not compatible with DNA intercalating viability dyes (propidium iodide, 7-AAD, DAPI).
• Use Invitrogen LIVE/DEAD Fixable Dead Cell stains with NovaFluor dyes.
• CellBlox Plus Blocking Buffer (Cat. No. C001T06F01) should be used for optimal staining. Add 5 µL per sample to antibody cocktails before combining with cells.

Target Information

IgE is one of five immunoglobulin classes in mammals. It consists of two heavy (epsilon) chains and two light chains. IgE mediates type I hypersensitivity reactions such as hay fever, asthma, hives, and anaphylactic shock by binding Fc receptors on basophils and mast cells. Crosslinking of IgE induces degranulation and release of mediators that drive allergic responses. IgE also contributes to parasite immunity.
