Description:
The monoclonal antibody IgE21 recognizes the immunoglobulin E (IgE) class of antibodies. Natural serum levels of IgE are low, representing a small fraction of total immunoglobulins. IgE mediates allergic responses by binding to mast cells and basophils via Fc epsilon receptor I, triggering release of histamine and other mediators. IgE is produced by terminally differentiated plasma B cells and basophils, and exists in both soluble and membrane-bound forms. IgE21 recognizes both forms.

Each product contains one vial of NovaFluor conjugate and one vial of CellBlox Plus Blocking Buffer.

Applications Reported:
This IgE21 antibody is reported for use in flow cytometric analysis.

Applications Tested:
Pre-titrated and tested by flow cytometric analysis of normal human peripheral blood cells. Use 5 µL (0.2 µg) per test. One test is defined as the amount of antibody required to stain a cell sample in 100 µL final volume. Cell number may range from 10⁵ to 10⁸ cells/test.

Important Notes:

• NovaFluor dyes are not compatible with DNA intercalating viability dyes (e.g., propidium iodide, 7-AAD, DAPI).
• Use Invitrogen LIVE/DEAD Fixable Dead Cell stains for compatibility.
• Ships with CellBlox Plus Blocking Buffer (Cat. No. C001T06F01), required for optimal staining.
• Add 5 µL of CellBlox Plus Blocking Buffer per sample, regardless of the number of NovaFluors in the panel.
• For single-color controls, use 5 µL per 100 µL of cell sample (10³–10⁸ cells).
• NovaFluor conjugates utilize Phiton™ technology for engineered fluorescent performance with reduced spectral spillover.

Target Information

IgE is one of five mammalian immunoglobulin classes. It consists of two epsilon heavy chains (with four Ig-like constant domains) and two light chains. IgE mediates type I hypersensitivity reactions (e.g., hay fever, asthma, hives, anaphylaxis) by binding Fc receptors on basophils and mast cells. Crosslinking of receptor-bound IgE induces degranulation and release of allergic mediators. IgE also contributes to immune defense against parasites.
