Product Specific Information

The monoclonal antibody IgE21 recognizes the immunoglobulin E (IgE) class of antibodies. Natural IgE levels in serum are low, comprising only a small portion of total immunoglobulin. IgE plays a role in allergy responses by binding to mast cells and basophils through the Fc epsilon receptor I, resulting in histamine release and other mediators. IgE is produced by terminally differentiated plasma B cells and basophils and exists in both soluble and membrane-bound forms. IgE21 recognizes both forms.

Each product contains:

• 1 vial of NovaFluor conjugate
• 1 vial of CellBlox Plus Blocking Buffer

Applications Reported: Flow cytometric analysis
Applications Tested: Pre-titrated and tested by flow cytometric analysis of normal human peripheral blood cells. Recommended use: 5 µL (0.2 µg) per test (100 µL final volume, 10⁵–10⁸ cells/test).

NovaFluor dyes are not compatible with DNA intercalating viability dyes (e.g., propidium iodide, 7-AAD, DAPI). Use Invitrogen LIVE/DEAD Fixable Dead Cell stains for viability assessment.

This NovaFluor conjugate ships with CellBlox Plus Blocking Buffer (Cat. No. C001T06F01), which contains formulation improvements for optimal staining. Add 5 µL per sample to antibody cocktails before combining with cells. For single-color controls, use 5 µL per 100 µL cell sample (10³–10⁸ cells).

NovaFluor conjugates utilize Phiton™ technology, based on novel nucleic acid dye structures engineered for optimized fluorescent signatures with minimal spillover and spread.

Target Information

IgE is one of five immunoglobulin classes in mammals. It consists of two heavy (epsilon) chains and two light chains. IgE plays a key role in type I hypersensitivity (allergic responses such as hay fever, asthma, hives, anaphylactic shock) by binding to Fc receptors on basophils and mast cells. Crosslinking of IgE induces degranulation, releasing mediators that promote allergic reactions. IgE also contributes to immunity against parasites.
