Product Specific Information

The monoclonal antibody IgE21 recognizes the immunoglobulin E (IgE) class of antibodies. IgE is present at low levels in serum and plays a key role in allergic responses by binding to mast cells and basophils through Fc epsilon receptor I, leading to histamine release. IgE is produced by plasma B cells and basophils, existing in both soluble and membrane-bound forms. IgE21 recognizes both forms.

Each product includes:

• 1 vial of NovaFluor conjugate
• 1 vial of CellBlox Plus Blocking Buffer

Applications Reported: Flow cytometric analysis
Applications Tested: Pre-titrated and tested by flow cytometry of normal human peripheral blood cells. Recommended use: 5 µL (0.2 µg) per test in 100 µL final volume. Cell count: 10⁵–10⁸ cells/test.

Note:
NovaFluor dyes are not compatible with DNA intercalating viability dyes (e.g., propidium iodide, 7-AAD, DAPI). Use Invitrogen LIVE/DEAD Fixable Dead Cell stains instead.

This product ships with CellBlox Plus Blocking Buffer (Cat. No. C001T06F01), which offers improved formulation over the original CellBlox. It is required for optimal staining with NovaFluor conjugates.

• Add 5 µL per sample to antibody cocktails before mixing with cells.
• For single-color controls: add 5 µL per 100 µL of cell sample (10³–10⁸ cells).

NovaFluor conjugates utilize Phiton™ technology with engineered nucleic acid dye structures for optimized fluorescent signatures, minimizing spillover and spread.

Target Information

IgE is one of five immunoglobulin classes in mammals, composed of two epsilon heavy chains and two light chains. It mediates type I hypersensitivity reactions (e.g., hay fever, asthma, hives, anaphylaxis) by binding to Fc receptors on basophils and mast cells. Crosslinking of IgE induces degranulation and release of mediators causing allergic symptoms. IgE also contributes to immunity against parasites.
