Product Specific Information

The monoclonal antibody IgE21 recognizes the immunoglobulin E (IgE) class of antibodies. IgE levels in serum are typically low, representing a small fraction of total immunoglobulins. IgE mediates allergic responses by binding to mast cells and basophils via Fc epsilon receptor I, leading to histamine release and other mediator secretion. IgE is produced by terminally differentiated plasma B cells and basophils and exists in both soluble and membrane-bound forms. IgE21 recognizes both.

Each product includes:

• 1 vial of NovaFluor conjugate
• 1 vial of CellBlox Plus Blocking Buffer

Applications Reported: Flow cytometric analysis
Applications Tested: Pre-titrated and tested on normal human peripheral blood cells.
Use 5 µL (0.2 µg) per test in 100 µL final volume. Suitable for 10⁵–10⁸ cells/test.

Important Notes:

• NovaFluor dyes are incompatible with DNA intercalating viability dyes (e.g., propidium iodide, 7-AAD, DAPI).
• Use Invitrogen LIVE/DEAD Fixable Dead Cell stains for viability assays.
• CellBlox Plus Blocking Buffer (Cat. No. C001T06F01) is required for optimal staining.
• Add 5 µL of CellBlox Plus Blocking Buffer per sample before combining with cells.
• For single-color controls, use 5 µL buffer per 100 µL of cell sample containing 10³–10⁸ cells.

NovaFluor conjugates utilize Phiton™ technology, employing nucleic acid dye structures engineered for optimal fluorescence with minimal spillover and spread.

Target Information

IgE is one of the five major immunoglobulin classes in mammals. It consists of two epsilon heavy chains and two light chains. IgE mediates type I hypersensitivity reactions such as hay fever, asthma, hives, and anaphylaxis by binding to Fc receptors on basophils and mast cells. Crosslinking of IgE on these cells induces degranulation and release of allergic mediators. IgE also contributes to immunity against parasites.
