Target Information
CREB (Cyclic AMP response element binding protein) is a 43 kDa basic/leucine zipper transcription factor that binds the cyclic AMP response element (CRE) and activates transcription in response to extracellular signals such as neurotransmitters, hormones, membrane depolarization, and growth/neurotrophic factors.
Activation of CREB depends on phosphorylation at serine 133. Phosphorylation occurs via p44/42 MAP kinase, p90RSK, p38 MAP kinase, and MSK1. While CREB binds DNA regardless of phosphorylation state, only the phosphorylated form functions as a transcription factor.
CREB binding protein (CBP), a transcriptional coactivator, interacts with CREB near serine 133. CREB participates in cellular processes including circadian rhythm regulation, adipocyte differentiation, and learning and memory.
In humans, the CREB gene is located on chromosome 2q. Dysfunction of CREB is associated with Alzheimer’s disease, histiocytoma, and soft tissue melanoma.
