Target Information
Connexin 43 (Cx43) is a gap junction protein that forms hexamers transported to the plasma membrane to create hemichannels. These hemichannels pair between adjacent cells to form gap junctions, essential for intercellular communication, development, and cell growth regulation.
Phosphorylation of Cx43 regulates assembly and function of gap junctions. Ser368 of Cx43 is phosphorylated by PKC upon activation by phorbol esters, reducing cell-to-cell communication. Src kinase also phosphorylates Cx43 to modulate junction function.
Connexin 43 is the major gap junction protein in the heart, playing a key role in synchronized contraction and embryonic development. Mutations in related genes such as GFAP are associated with Charcot-Marie-Tooth disease, oculodentodigital dysplasia, and heart malformations.
