Product Specific Information

This antibody detects endogenous levels of Progesterone Receptor only when phosphorylated at Ser345.

Target Information

The progesterone receptor (PR) is a member of the steroid family of nuclear receptors. PR mediates the physiological effects of progesterone, which plays a central role in reproductive events associated with the establishment and maintenance of pregnancy.
PR exists as two isoforms: a 94 kDa protein (Form A) and a 120 kDa protein (Form B), resulting from alternative translation initiation sites.

  • PR-B: Transcriptionally active form responsible for endometrial maintenance, pregnancy maintenance, and inhibition of ovulation.
  • PR-A: Identical to PR-B except for a 165 amino acid deletion at the N-terminus, exposing a 140 amino acid inhibitory domain that represses steroid hormone transcriptional activity.

In its inactive state, the progesterone receptor forms a multiprotein complex including heat shock proteins and immunophilins. Upon hormone binding, conformational changes allow dimerization and binding to progesterone response elements (PRE), leading to transcription activation.
A null mutation in the PGR gene can result in reproductive abnormalities.
