Product Specific Information

The CBRM1/5 monoclonal antibody reacts with an activation-specific epitope of human Mac-1. It binds to a subset of Mac-1 molecules on neutrophils and monocytes after stimulation but does not recognize resting myeloid cells. Mac-1 participates in adhesive cell interactions through its ligands.
The epitope recognized by CBRM1/5 localizes to the I domain on the alpha chain of Mac-1, near the ligand binding site. This antibody blocks Mac-1-dependent adhesion to fibrinogen and ICAM-1 and inhibits chemoattractant-stimulated adhesion of eosinophils to ICAM-1.
Low-level activation may occur during processing of freshly drawn blood, leading to some binding in unstimulated samples; however, higher levels are observed in activated samples.

Applications Reported: Flow cytometry, immunoprecipitation, and blocking of CD11b function.
For functional assays, use the Functional Grade purified CBRM1/5 (Product #16-0113).

Applications Tested: Tested by flow cytometric analysis of resting and activated normal human peripheral blood cells. Recommended ≤1 µg/test (100 µL final volume). Cell numbers may vary (10⁵–10⁸ cells/test). Antibody should be titrated for optimal performance.

Target Information

CD11b (integrin alpha-M, ITGAM, integrin alpha-X, ITGAX) is a 165 kDa adhesion molecule that associates non-covalently with integrin beta-2 (CD18). The CD11b/CD18 heterodimer (Mac-1, CR3) functions as a receptor for ICAM-1, ICAM-2, ICAM-4, CD14, CD50, CD23, heparin, iC3b, fibrinogen, and Factor X.
It is expressed on monocytes/macrophages, granulocytes, activated lymphocytes, subsets of NK and dendritic cells, and microglia. CD11b expression increases during monocyte maturation and varies among tissue macrophages.
Associated diseases include systemic lupus erythematosus and ITGAM-related susceptibility to autoimmune disorders.
