Product Specific Information

The HIT8a monoclonal antibody reacts with the human CD8a molecule, a 32–34 kDa cell surface receptor expressed as either a heterodimer with CD8 beta chain (CD8α/β) or a homodimer (CD8α/α). CD8a is found on most thymocytes and subsets of mature T cells and NK cells. It binds to MHC class I molecules and, through association with protein tyrosine kinase p56lck, plays a role in T cell development and activation.

Applications Tested:

• Tested by flow cytometric analysis of normal human peripheral blood cells.
• Use ≤ 0.5 µg per test (defined as antibody amount staining 10⁵–10⁸ cells in 100 µL volume).

Target Information

CD8 (Cluster of Differentiation 8) is a type I transmembrane glycoprotein of the immunoglobulin family. It functions as a co-receptor with T cell receptor (TCR) for MHC class I molecules, facilitating T cell activation and differentiation. CD8 is typically expressed as a disulfide-linked heterodimer (CD8α/β) on cytotoxic T cells and as a homodimer (CD8α/α) on subsets of T cells, NK cells, monocytes, and dendritic cells.

Binding of MHC-I/peptide complexes to CD8 recruits Lck kinase, triggering lymphokine production, cell motility, and cytotoxic T lymphocyte (CTL) activation. Activated CTLs are critical for pathogen and tumor cell clearance. Differentiation of naïve CD8⁺ T cells into CTLs is enhanced by IL-2, IL-12, and TGF-β1.
