Product Specific Information

The 53-6.7 monoclonal antibody reacts with mouse CD8a, a 32–34 kDa cell surface receptor expressed as either a heterodimer (CD8αβ) or homodimer (CD8αα). CD8 is found on thymocytes, mature αβ T cells, γδ T cells, intestinal intraepithelial lymphocytes, and dendritic cells. It binds to MHC class I and associates with tyrosine kinase p56^lck, playing a role in T cell development and activation.

Applications Reported:
Used for flow cytometry, immunoprecipitation, and frozen tissue immunohistology. Also applied in T cell activation, CD8+ cell isolation/depletion, and ligand blocking.
For functional assays, use Functional Grade purified 53-6.7 (Product #16-0081).

Applications Tested:
Validated by flow cytometric analysis of mouse thymocytes and splenocytes at ≤0.25 µg/test (100 µL final volume). Optimal performance requires empirical titration.

Target Information

CD8 (Cluster of Differentiation 8) is a type I transmembrane glycoprotein in the immunoglobulin receptor family. It acts as a co-receptor with TCR for MHC class I molecules, facilitating T cell signal transduction, differentiation, and activation. CD8 is expressed as CD8αβ heterodimers on T cells and as CD8αα homodimers on subsets of T cells, NK cells, and dendritic cells. Engagement with MHC-I/peptide complexes recruits Lck kinase, inducing cytokine production and cytotoxic T lymphocyte (CTL) activation. CTLs are essential for pathogen and tumor clearance and are enhanced by IL-2, IL-12, and TGF-β1.
