Product Specific Information

The JES5-16E3 antibody reacts with mouse interleukin-10 (IL-10), an ~18 kDa cytokine known as Cytokine Synthesis Inhibitory Factor (CSIF).
In mice, IL-10 is produced mainly by Th2 cells, B1 cells, macrophages, and keratinocytes. It inhibits Th1 cytokine synthesis, suppresses T-cell proliferation, and acts as a co-stimulatory signal for mast cells and developing thymocytes.

Applications Reported:
Functional Grade Biotin anti-mouse IL-10 antibody (clone JES5-16E3) is used as the in vivo capture antibody for the In Vivo Cytokine Capture Assay (IVCCA), which increases cytokine measurement sensitivity by extending their in vivo half-lives.

Applications Tested:
The Functional Grade Biotin JES5-16E3 antibody is LAL-tested and endotoxin-free. It has been used as the detection antibody in sandwich ELISA for mouse IL-10, in combination with JES5-2A5 (14-7102) for capture and recombinant mouse IL-10 (14-8101) as standard.
Recommended ELISA detection concentration: 0.5–2 µg/mL.
Standard curve range: 4000 pg/mL – 30 pg/mL.

Endotoxin: Less than 0.05 ng/µg antibody (LAL assay).
Storage and Handling: Use in a sterile environment.


Target Information

IL-10 is a cytokine primarily produced by monocytes and lymphocytes. It regulates immune responses and inflammation by down-regulating Th1 cytokines, MHC class II antigens, and costimulatory molecules on macrophages.
IL-10 enhances B cell survival, proliferation, and antibody production, and inhibits NF-κB activity. It plays a key role in JAK-STAT signaling regulation.
Knockout studies in mice show IL-10 is essential for intestinal immune regulation. Mutations in IL10 are linked to increased susceptibility to HIV-1 infection and rheumatoid arthritis.
Associated diseases include Graft-Versus-Host Disease and Human Immunodeficiency Virus Type 1.
