Product Specific Information

The 6H6 monoclonal antibody reacts with human CD123, the alpha chain of the IL-3 receptor. This 60–70 kDa transmembrane protein binds IL-3 with low affinity by itself, and with CD131 forms a high-affinity receptor complex. CD123 is expressed by myeloid precursors, macrophages, dendritic cells, mast cells, basophils, and megakaryocytes.

• Applications Reported: Flow cytometric analysis
• Applications Tested: Pre-diluted and validated for flow cytometry using normal human peripheral blood cells. Recommended use: 5 µL (0.25 µg) per 100 µL cell sample (10⁵–10⁸ cells/test).

Fluorochrome Information:
Brilliant Ultra Violet™ 737 (BUV737) emits at 732 nm and is designed for use with ultraviolet (355 nm) lasers. Ensure your cytometer supports this fluorochrome.

Buffer Recommendations:
When using multiple polymer dye-conjugated antibodies (e.g., Super Bright, Brilliant Violet™, Brilliant Ultra Violet™), use Super Bright Complete Staining Buffer (SB-4401) or Brilliant Stain Buffer (00-4409) to minimize non-specific interactions.

Light Sensitivity:
Protect from light to prevent photo-induced oxidation.

Fixation Compatibility:
Samples can be stored up to 3 days in the dark at 4°C using IC Fixation Buffer (00-8222) or 1-step Fix/Lyse Solution (00-5333). Brilliant Ultra Violet™ 737 is compatible with short-term methanol fixation (<1 hour).

Target Information

IL-3Ra (Interleukin-3 receptor subunit alpha) belongs to the type I cytokine receptor family, forming a heterodimer with the common beta (CDw131) subunit. It mediates IL-3 signal transduction and becomes activated upon IL-3-induced heterodimerization. IL-3Ra is overexpressed in acute myeloid leukemia (AML) blasts compared to normal hematopoietic cells, representing a potential therapeutic target.
