Product Specific Information
This antibody detects endogenous levels of SHP-1 only when phosphorylated at Tyrosine 536.
Target Information
SHP-1 (PTPN6) is a non-receptor protein tyrosine phosphatase expressed primarily in hematopoietic cells. It consists of two SH2 domains, a tyrosine phosphatase catalytic domain, and a carboxy-terminal regulatory domain. SHP-1 downregulates several tyrosine kinase-regulated pathways by dephosphorylating target proteins. In hematopoietic cells, SHP-1 binds to tyrosine-phosphorylated erythropoietin receptors (EpoR) to negatively regulate growth. Overexpression in epithelial cells leads to dephosphorylation of Ros receptor tyrosine kinase, reducing Ros-dependent proliferation and transformation. In myeloid cells, SHP-1 associates with IL-3R beta chain to downregulate IL-3-induced tyrosine phosphorylation and cell proliferation.
