Product Specific Information

PA1-053 detects leptin (OB-R) from human, mouse, and rat tissues.
It has been successfully used in Western blot procedures.
By Western blot, this antibody detects ~100 kDa and ~125 kDa proteins representing the short and long forms of OB-R from mouse brain extract.
It also detects two smaller, unrelated proteins (~31 kDa and ~35 kDa) in mouse liver extract that cannot be competed with the peptide.

The immunogen is a synthetic peptide corresponding to residues K(577)–A(594) of the extracellular domain of rat OB-R, completely conserved in mouse OB-R.
The PA1-053 immunizing peptide (Cat. # PEP-014) is available for use in neutralization and control experiments.

Target Information

The leptin (OB-R) receptor belongs to the cytokine receptor superfamily and plays a crucial role in body weight homeostasis and energy balance.
Activation by leptin triggers downstream signaling via the JAK/STAT pathway.
Leptin can homo-dimerize OB-R extracellular domains, initiating intracellular signaling upon dimerization.

Several OB-R isoforms with varying lengths and signaling capabilities exist.
RNA transcripts for the OB-R long form are found in the choroid plexus and hypothalamus, key regions for satiety and energy control.
Mutations in mouse OB-R (db) or leptin (ob) genes lead to early-onset obesity and metabolic abnormalities such as hyperinsulinemia, insulin resistance, hyperglycemia, and increased adipose tissue.
