Product Specific Information
This 4B4 (4B4-1) monoclonal antibody reacts with human CD137 (also known as 4-1BB or TNFRSF9), an inducible member of the TNFR family of costimulatory molecules expressed on T cells, NK cells, dendritic cells, granulocytes, and mast cells.
CD137 signaling recruits TRAF1 and TRAF2, playing roles in T cell activation, survival of activated and CD8 memory T cells, and suppression of myelopoiesis and dendritic cell development.
Stimulation of CD137 promotes expansion of CD4+CD25+ regulatory T cells ex vivo.
Its ligand, 4-1BBL, is expressed on activated macrophages, mature B cells, hematopoietic stem cells, and myeloid progenitor cells.
Applications Reported: Flow cytometric analysis.
Applications Tested: Pre-diluted and tested by flow cytometry using stimulated normal human peripheral blood cells. Recommended usage: 5 µL (0.25 µg) per test (100 µL final volume).
Cell number should be empirically determined (10⁵–10⁸ cells/test).
Fluorochrome Information:
Alexa Fluor 700 emits at 723 nm and is excited by red laser (633–647 nm).
Recommended optics: 685 LP mirror and 710/20 band pass filter.

Target Information
CD137 (TNFRSF9, 4-1BB) is an inducible costimulatory molecule mainly expressed on activated T cells.
Its ligand, 4-1BBL, is found on activated macrophages, mature B cells, hematopoietic stem cells, and myeloid progenitor cells.
CD137 signaling supports survival of activated and memory T cells and promotes T cell clonal expansion, while suppressing myelopoiesis and dendritic cell development.
Triggered CD137 induces cytokine release regulating peripheral monocyte survival.
Soluble CD137 may act as a negative regulator for some immune responses.
