Product Specific Information

The eBioGHI/61 monoclonal antibody reacts with human CD163 (Haemoglobin Scavenger Receptor, M130), a member of the macrophage scavenger receptor family. CD163 is expressed by tissue macrophages, circulating blood monocytes, and possibly by hematopoietic progenitor cells. It functions as a high-affinity scavenger receptor for the hemoglobin-haptoglobin complex, facilitating clearance of hemoglobin from the bloodstream.

Upon ligand binding, CD163 triggers signaling leading to IL-6 and IL-10 production. Expression of CD163 is induced by anti-inflammatory cytokines (IL-6, IL-10) and downregulated by proinflammatory stimuli (LPS, TNF-α), which can cause shedding of soluble CD163.

Applications Reported: Flow cytometric analysis.
Applications Tested: Pre-diluted and tested on normal human peripheral blood cells. Use at 5 µL (0.25 µg) per test in 100 µL final volume. Suitable for 10⁵–10⁸ cells/test.

Target Information

CD163 (M130 antigen, Ber-Mac3, Ki-M8, SM4) is a 130 kDa membrane glycoprotein of the scavenger receptor cysteine-rich superfamily. It serves as a receptor for the hemoglobin-haptoglobin complex, protecting tissues from oxidative damage and aiding in iron recycling.

CD163 is expressed on monocytes and macrophages, particularly during late inflammation. It is found on CD14⁺ and CD64⁺ monocytes, with higher expression on CD16⁺ subsets. Expression is upregulated by IL-10, IL-6, and dexamethasone, and downregulated by LPS or PMA through shedding into plasma.

Binding of CD163 to hemoglobin/haptoglobin complexes is calcium- and pH-dependent, showing higher affinity for multimeric haptoglobin (HP1F phenotype). It also induces intracellular signaling involving tyrosine kinase-dependent calcium mobilization, inositol triphosphate production, and secretion of IL-6 and CSF1.
