Product Specific Information

The eBioRDR5 monoclonal antibody reacts with human CD127 (Interleukin-7 Receptor alpha). CD127 complexes with CD132 (common gamma chain) to form the IL-7 receptor (IL-7R). CD127 is a type I glycoprotein (75–80 kDa) expressed on immature B cells, thymocytes, and most mature T cells, with transient down-regulation upon activation.

IL-7 binding triggers signal transduction via tyrosine kinase pathways, including Jak/STAT. IL-7 is essential for lymphocyte development and T-cell homeostasis. IL-7R signaling also regulates TCR locus rearrangement in γδ T cells.

CD127 expression is down-regulated on CD4⁺CD25⁺ regulatory T cells (Tregs). Co-expression of CD4 and CD25 identifies Tregs, but inclusion of CD127 down-regulation enriches for Foxp3⁺ suppressive Tregs. Binding of eBioRDR5 to PBMCs is blocked by recombinant human IL-7 (Product #14-8079). The antibody cross-reacts with rhesus CD127.

This antibody is pre-titrated and validated for flow cytometry on normal human PBMCs. Recommended use: 5 µL (0.125 µg)/test in 100 µL final volume. Suitable for 10⁵–10⁸ cells/test.

Alexa Fluor® 700 emits at 723 nm and is excited by a 633 nm red laser. Recommended detection: 685 LP mirror and 710/20 filter. Ensure instrument compatibility.
Filtration: 0.2 µm post-manufacturing filtered.

Target Information

CD127 (Interleukin-7 receptor alpha, IL7Rα) is a glycoprotein involved in lymphopoiesis regulation. The active IL7R complex is an alpha/gamma chain heterodimer. The gamma(c) chain activates signal transduction, while the alpha chain mediates specific signaling events. CD127 promotes proliferation of precursor B cells, thymocytes, T cell progenitors, and mature CD4⁺/CD8⁺ T cells.

Dysfunction of CD127 is associated with severe combined immunodeficiency (SCID) and T cell negative/B cell negative/NK positive SCID.
